Background
Sugemalimab Biosimilar (Research Grade, ICH5278) is an unconjugated, non-therapeutic analog of the originator anti-PD-L1 antibody sugemalimab, produced for research use only. It is built to engage the same molecular target as the reference drug, programmed death-ligand 1 (PD-L1/CD274), and is supplied as a full-length human IgG4-lambda immunoglobulin matching the reported isotype of the originator. As a research-grade biosimilar it is intended for in-vitro and functional studies rather than any clinical or diagnostic application. Sugemalimab is a fully human anti-PD-L1 monoclonal that blocks the PD-1/PD-L1 checkpoint axis; the originator has been developed in oncology, including non-small cell lung cancer. This analog lets investigators study PD-L1 engagement, ligand-receptor blockade, and downstream immune readouts without sourcing clinical material. It is offered at low endotoxin levels suitable for sensitive cell-based assays and is available in bulk milligram-to-gram quantities as an in-vivo/assay standard. Note the supplied card labels the target as PD-1; the drug and UniProt entry correspond to PD-L1, so the material should be understood as anti-PD-L1.
PD-L1 (CD274, B7-H1) is the primary ligand for the inhibitory receptor PD-1 and is the actual target of sugemalimab (UniProt Q15116). It is a type I transmembrane glycoprotein of the B7 family expressed on tumor cells, antigen-presenting cells, and many stromal and epithelial tissues, often upregulated by interferon-gamma and other inflammatory signals. Engagement of PD-L1 with PD-1 on activated T cells delivers an inhibitory signal that dampens T-cell receptor signaling, reduces proliferation and cytokine production, and promotes T-cell exhaustion, a key mechanism of tumor immune evasion. PD-L1 also binds CD80 (B7-1). Antibodies that block PD-L1 free PD-1-bearing effector T cells from this brake, restoring cytotoxic anti-tumor responses. Because the checkpoint acts at the ligand side, anti-PD-L1 reagents are widely used to interrogate immune-suppression pathways in vitro.
