Background
This product is an unconjugated, non-therapeutic recombinant analog of nofazinlimab, an anti-PD-1 monoclonal antibody, supplied strictly for research use only. It is built around the same target, human programmed cell death protein 1 (PD-1/PDCD1, UniProt Q15116), and reproduces the human IgG4 kappa framework of the originator so that investigators can study PD-1 pathway biology and antibody function without a clinical-grade drug. It is not the clinical product and is not intended for human or veterinary use. As a research-grade reagent it is offered at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities, making it well suited as an isotype-matched, target-defined tool for binding and blockade assays, positive and comparator controls in checkpoint screens, ADCC/effector or ADC-development workflows, and preclinical in-vitro and ex-vivo model systems. The IgG4 backbone is relevant because it minimizes effector engagement, mirroring the checkpoint-blocking design intent of the parent molecule.
PD-1 (programmed cell death protein 1; gene PDCD1; UniProt Q15116) is a type I transmembrane immunoinhibitory receptor of the CD28/immunoglobulin superfamily, expressed on activated T cells, B cells, natural killer cells, and other immune subsets. Its cytoplasmic tail carries an immunoreceptor tyrosine-based inhibitory motif (ITIM) and switch motif (ITSM) that, upon ligand engagement, recruit the phosphatase SHP-2 to dampen TCR and CD28 signaling. Its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2) are expressed on antigen-presenting cells, many tumor cells, and inflamed tissue. Under normal conditions this axis maintains peripheral tolerance and limits immunopathology, but tumors exploit it to suppress cytotoxic T-cell responses. Blocking PD-1 engagement can restore effector T-cell activity, which underpins the interest in PD-1 as a checkpoint target.
