Background
This product is a research-grade biosimilar of zimberelimab, an anti-PD-1 antibody. It is supplied as an unconjugated, non-therapeutic analog of the originator molecule and is intended strictly for research use only; it is not a clinical drug and is not for human or veterinary use. The antibody is built around the programmed cell death protein 1 (PD-1) target and is expressed as a human IgG4 (lambda) format, mirroring the isotype of the reference molecule. As with other IgG4-based checkpoint biosimilars, it recapitulates the antigen-binding properties of the originator while carrying the stabilized, low-effector-function characteristics associated with the IgG4 subclass. It is manufactured to research-grade endotoxin specifications (typically less than 1 EU/mg), and is available in bulk milligram-to-gram quantities suitable for in-vitro assays, screening cascades, and use as a reference or comparator reagent. It provides a cost-effective, consistently sourced alternative to therapeutic material for investigators studying PD-1 biology, checkpoint blockade mechanisms, and assay development, where a well-characterized anti-PD-1 binder is required. PD-1 (programmed cell death protein 1, gene PDCD1) is an inhibitory immune checkpoint receptor expressed on activated T cells, B cells, and myeloid cells. It is a type I transmembrane protein of the immunoglobulin superfamily bearing intracellular ITIM and ITSM motifs. Engagement by its ligands PD-L1 and PD-L2 recruits the phosphatase SHP-2, which dampens TCR and CD28 signaling, attenuating T-cell activation, proliferation, and cytokine production. This pathway maintains peripheral tolerance and limits immunopathology, but tumors and chronic infections exploit it to drive T-cell exhaustion and immune evasion. Antibodies that block the PD-1/PD-L1 axis restore effector T-cell function and underpin a major class of cancer immunotherapies. PD-1 is therefore a central node in studies of tumor immunology, checkpoint blockade, and T-cell dysfunction.
