Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-PD-1 antibody ezabenlimab, supplied for research use only. It reproduces a human IgG4-kappa format directed against human PD-1 (PDCD1, UniProt Q15116) and is intended to block the PD-1/PD-L1 and PD-1/PD-L2 interaction in laboratory settings. It is not the clinical drug and is not for human or veterinary use. The recombinant format makes it useful as a defined, lot-consistent reagent for PD-1 checkpoint research: as a positive control or benchmarking tool in PD-1/PD-L1 blockade assays, as a capture or detection reagent in binding and epitope studies, and as a comparator in biosimilarity or ADCC/ADC-development workflows. Because it is offered unconjugated, users can label or immobilise it for their own assay formats. It is manufactured to low-endotoxin research grade (typically under 1 EU/mg, with an ultra-low under 0.5 EU/mg option) and is available in bulk milligram-to-gram quantities to support screening campaigns, preclinical model work, and reagent standardisation across experiments. PD-1 (programmed cell death protein 1; gene PDCD1, UniProt Q15116) is a type I transmembrane immune checkpoint receptor of the CD28/immunoglobulin superfamily, expressed on activated T cells and, to varying degrees, on B cells, NK cells, and myeloid subsets. Its cytoplasmic tail carries ITIM and ITSM motifs that, upon ligand engagement, recruit the phosphatase SHP-2 to dampen TCR and CD28 signalling, promoting T-cell exhaustion and peripheral tolerance. PD-1 has two ligands, PD-L1 (CD274) and PD-L2 (CD273), which are frequently upregulated in the tumour microenvironment, allowing tumours to evade immune attack. Antibodies that block PD-1 from binding PD-L1/PD-L2 relieve this inhibitory signal and restore effector T-cell activity, the mechanistic basis of PD-1 checkpoint blockade in immuno-oncology research.
