Background
This product is an unconjugated, non-therapeutic recombinant analog of pidilizumab, an antibody historically developed against the T-cell checkpoint receptor PD-1 (PDCD1). It is supplied as a research-grade reagent for research use only and is not the clinical drug, nor is it intended for human or veterinary use. Built as a human IgG1/kappa molecule directed at the PD-1 antigen, it is useful as a defined, sequence-consistent tool for in-vitro and preclinical work where a reproducible anti-checkpoint binder is needed: as a positive or reference control in binding and blockade assays, as a benchmarking comparator alongside other anti-PD-1 analogs, and as a starting reagent for ADCC or antibody-engineering and ADC-development workflows. Low-endotoxin grades (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) support cell-based and in-vivo-standard applications where endotoxin would confound readouts. Available unconjugated and in bulk milligram-to-gram quantities to support assay development, screening campaigns, and preclinical model studies.
PD-1 (PDCD1) is an inhibitory immune-checkpoint receptor of the CD28/CTLA-4 family, expressed on activated T cells, B cells, NK cells, and other immune subsets. It is a type I transmembrane glycoprotein with a single IgV-like extracellular domain and a cytoplasmic tail bearing ITIM and ITSM motifs. On engagement by its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2), PD-1 recruits the phosphatase SHP-2, dampening TCR and CD28 signaling and attenuating T-cell proliferation, cytokine production, and effector function. This pathway maintains peripheral tolerance and limits immunopathology, but tumors and chronically infected tissues exploit it to drive T-cell exhaustion. Blocking the PD-1/PD-L1 axis restores T-cell activity, the rationale behind checkpoint-inhibitor immunotherapy.
