Tamgiblimab Biosimilar (Research Grade) is an unconjugated, non-therapeutic research analog of the anti-TIGIT antibody tamgiblimab (originator code IBI939, Innovent Biologics), a fully human IgG4-kappa monoclonal originally developed as a checkpoint immunotherapy for solid tumors, typically explored in combination with PD-1/PD-L1 blockade. This product is built to recapitulate the binding characteristics of the originator against its target, human TIGIT (T-cell immunoreceptor with immunoglobulin and ITIM domains), and is supplied strictly for research use only. It is not a clinical drug and is not intended for therapeutic, diagnostic, or human use. Matching the originator, it carries a human IgG4-kappa framework, which limits Fc effector engagement relative to IgG1-based checkpoint agents. The biosimilar is intended as a consistent, well-characterized reagent for studies of TIGIT biology, ligand-blockade, and checkpoint-pathway crosstalk, and as a reference or control antibody in assay development. As a human-targeted molecule, it is most suited to in-vitro and functional work rather than mouse in-vivo dosing. Available in bulk quantities with low endotoxin research-grade specifications. TIGIT is an inhibitory immune checkpoint receptor expressed on activated T cells, regulatory T cells (Tregs), and natural killer (NK) cells. It binds the ligands CD155 (PVR) and CD112 (PVRL2/nectin-2), which are frequently overexpressed on tumor cells and antigen-presenting cells. Engagement of TIGIT delivers inhibitory signals that dampen T-cell and NK-cell activation and promote functional exhaustion of tumor-infiltrating lymphocytes. TIGIT also competes with the co-stimulatory receptor CD226 (DNAM-1) for the same ligands, further tilting the balance toward immunosuppression, and it can enhance Treg suppressive activity. Because CD155 is upregulated in many malignancies, the TIGIT axis is a prominent target in cancer immunotherapy, often pursued alongside PD-1/PD-L1 blockade. Antibodies that block TIGIT-ligand interactions aim to restore cytotoxic T-cell and NK-cell function within the tumor microenvironment.
