DKK1 (Dickkopf-related protein 1, UniProt O94907) is a secreted glycoprotein and a principal antagonist of the canonical Wnt/beta-catenin signaling pathway. It acts primarily by binding the Wnt co-receptor LRP5/LRP6, and in concert with Kremen proteins promotes receptor internalization, thereby dampening Wnt-driven transcription. Through this activity DKK1 helps regulate embryonic patterning, stem-cell maintenance, and bone remodeling, where it restrains osteoblast activity. DKK1 is frequently dysregulated in cancer, where it can influence tumor cell behavior and the surrounding immune microenvironment, and elevated levels have been associated with poor outcomes in several tumor types. Its role at the interface of Wnt signaling, bone metabolism, and tumor-immune crosstalk makes DKK1 an attractive target for antibody-based intervention, which is the biological rationale underlying sirexatamab-class molecules that neutralize DKK1 to modulate downstream Wnt and immune effects.
