Background

This product is an unconjugated, non-therapeutic recombinant analog of the anti-PD-1 antibody enlonstobart, engineered around the human PD-1 (PDCD1) target and supplied strictly for research use only. It is a human IgG4-kappa molecule that reproduces the antigen-binding behavior of the originator drug, allowing researchers to study PD-1 engagement without using clinical material. It is not the therapeutic product and is not intended for human or veterinary use. The analog is useful as a defined, reproducible reagent for in-vitro binding and blocking assays, as a positive control or benchmarking tool against other anti-PD-1 reagents, for developing and validating assays (ELISA, flow cytometry, SPR/BLI affinity work), and for antibody-engineering and ADC/ADCC development workflows. It is available in bulk milligram-to-gram quantities with low-endotoxin research grade (<1 EU/mg) and ultra-low endotoxin (<0.5 EU/mg) options suitable for sensitive cell-based and preclinical studies. Consistent lot-to-lot performance makes it well suited to comparative and screening experiments where a stable reference standard is required. PD-1 (programmed cell death protein 1; gene PDCD1; UniProt Q15116) is an inhibitory immune checkpoint receptor of the CD28/B7 family, expressed on activated T cells, B cells, NK cells, and myeloid subsets. It carries an immunoreceptor tyrosine-based inhibitory motif (ITIM) and switch motif (ITSM) in its cytoplasmic tail. Engagement by its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2) recruits the phosphatase SHP-2, dampening TCR and CD28 signaling and attenuating T-cell activation, proliferation, and cytokine output. This pathway maintains peripheral tolerance and limits immunopathology, but tumors and chronic infections exploit it to drive T-cell exhaustion. Blocking the PD-1/PD-L1 axis restores effector function, the basis of checkpoint-inhibitor immunotherapy. PD-1 is a central node in studies of T-cell exhaustion, tumor immune evasion, and combination immunotherapy.
