ICH5417 is a research-grade biosimilar of budigalimab, supplied as an unconjugated, non-therapeutic analog of the originator anti-PD-1 antibody. It is built around the PD-1 (PDCD1) target and produced as a human IgG1, kappa immunoglobulin for research use only; it is not the clinical drug and is not intended for diagnostic or therapeutic use. Budigalimab is an investigational humanized monoclonal antibody directed against programmed cell death protein 1, developed as an immune-checkpoint inhibitor and studied across several solid-tumor settings. This biosimilar reproduces the target-binding specificity of the originator so that laboratories can study PD-1 engagement, blockade, and downstream signaling without sourcing clinical material. It is manufactured to research-grade specifications with low endotoxin (typically less than 1 EU/mg), and is available in bulk milligram-to-gram quantities suitable for repeated in-vitro assays, method development, and reference/comparator work. Because it is a defined recombinant reagent rather than a hybridoma product, it is characterized by its target and originator identity rather than by a hybridoma clone designation. PD-1 (programmed cell death protein 1), encoded by PDCD1 and corresponding to UniProt Q15116, is an inhibitory immune checkpoint receptor of the CD28/CTLA-4 family expressed on activated T cells, B cells, NK cells, and other lymphocytes. Its cytoplasmic tail carries ITIM and ITSM motifs that recruit the phosphatase SHP-2 upon ligand engagement, dampening TCR and costimulatory signaling. PD-1 binds two ligands, PD-L1 (CD274) and PD-L2 (PDCD1LG2), which are frequently upregulated in the tumor microenvironment and on antigen-presenting cells. Sustained PD-1 signaling contributes to T-cell exhaustion and peripheral tolerance, limiting antitumor and antiviral responses. Antibodies that block the PD-1/PD-L1 axis restore effector T-cell function, the pharmacological basis of checkpoint-inhibitor immunotherapy. PD-1 remains a central node for studying tumor immune evasion, exhaustion phenotypes, and combination immunotherapy strategies.
