Background

This is a research-grade biosimilar of Pradusinstobart: an unconjugated, non-therapeutic recombinant human IgG4-kappa antibody analog built around the immune checkpoint receptor PD-1 (PDCD1, UniProt Q15116). It reproduces the antigen-binding specificity of the originator anti-PD-1 molecule and is supplied for research use only. It is not the clinical drug and is not intended for human or veterinary use. The format makes it useful as a defined, well-characterized tool for PD-1/PD-L1 pathway work: as a binding and blockade reagent in receptor-ligand assays, as a positive control or benchmarking standard against other anti-PD-1 clones, as a capture or detection reagent, and as a scaffold for ADCC/ADC-development and engineering studies. The IgG4 backbone (typically stabilized) recapitulates the reduced effector-function profile expected of checkpoint-blocking antibodies. Material is offered at research grade with low endotoxin (<1 EU/mg standard; <0.5 EU/mg ultra-low options), enabling sensitive cell-based and in-vitro immunology assays, and is available in bulk milligram-to-gram quantities to support preclinical and assay-development pipelines requiring lot-consistent, animal-origin-free recombinant protein. PD-1 (Programmed cell death protein 1; gene PDCD1; UniProt Q15116; also CD279) is a type I transmembrane immunoinhibitory receptor of the CD28/B7 immunoglobulin superfamily. It is expressed on activated T cells, B cells, NK cells, and other immune subsets, and is upregulated with chronic antigen exposure. Its cytoplasmic tail carries an ITIM and an ITSM; engagement by its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2) recruits the phosphatase SHP-2, which dephosphorylates TCR and CD28 signaling components, dampening T-cell activation, proliferation, and cytokine output. This axis maintains peripheral tolerance and limits immunopathology, but tumors and chronically infected tissues exploit it to drive T-cell exhaustion. Blocking PD-1 or its ligands restores effector function, the mechanistic basis of checkpoint-inhibitor immunotherapy.
