Background
This product is an unconjugated, non-therapeutic recombinant analog of gilvetmab, an anti-PD-1 (PDCD1) monoclonal antibody, supplied for research use only. It is built around the PD-1 target as a research-grade tool and is not the clinical or veterinary drug; it is not intended for human or animal treatment. As a functional analog it is useful as a positive control and reference reagent in PD-1 binding, blockade, and checkpoint-modulation assays, for characterizing PD-1/PD-L1 pathway biology, for benchmarking other anti-PD-1 reagents, and as a building block in ADCC or antibody-engineering and ADC-development workflows. The antibody is offered unconjugated so users can apply their own labels or secondary detection. Formats are typically low-endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and available in bulk milligram-to-gram quantities to support in-vitro assays and preclinical model work. Because it is a research reagent rather than a therapeutic, lot-appropriate validation for the intended application is recommended. PD-1 (programmed cell death protein 1), encoded by PDCD1, is a type I transmembrane immunoinhibitory receptor of the CD28/immunoglobulin superfamily. It is expressed on activated T cells, B cells, natural killer cells, and other immune subsets, and is markedly upregulated on chronically stimulated or exhausted T cells. Its cytoplasmic tail carries ITIM and ITSM motifs that, upon ligand engagement, recruit the phosphatase SHP-2 to dampen TCR and CD28 signaling. PD-1 binds two ligands, PD-L1 (CD274) and PD-L2 (PDCD1LG2), which are expressed on antigen-presenting cells and, aberrantly, on many tumors. Ligation delivers a coinhibitory signal that reduces T-cell proliferation, cytokine production, and cytotoxicity, contributing to peripheral tolerance and to tumor immune evasion. Blocking the PD-1/PD-L1 axis restores effector T-cell function, the mechanistic basis of checkpoint-inhibitor immunotherapy.
