Background
This is a research-grade biosimilar of pimivalimab (JTX-4014), an anti-PD-1 antibody. It is an unconjugated, non-therapeutic recombinant analog built around the PD-1 target, produced as a fully human IgG4-kappa immunoglobulin. It is intended strictly for research use only (RUO) and is not the clinical drug; it is not for human or veterinary use. Because it reproduces the antigen-binding behavior of the originator against human PD-1, it is useful as a defined, reproducible reagent for in-vitro and preclinical work: as a positive control or benchmarking standard alongside other checkpoint blockers, in receptor-ligand binding and blockade assays, in reporter and functional T-cell assays, and as a starting antibody for ADCC/ADC or bispecific development workflows. The material is offered at low-endotoxin research grade (typically less than 1 EU/mg, with ultra-low options under 0.5 EU/mg) and available in bulk milligram-to-gram quantities, supporting assay standardization, in-vitro model systems, and larger preclinical studies where lot-to-lot consistency and cost per milligram matter.
PD-1 (programmed cell death protein 1; gene PDCD1, UniProt Q15116) is a type I transmembrane co-inhibitory receptor of the CD28/CTLA-4 immunoglobulin superfamily, expressed on activated T cells and also on B cells, NK cells, and some myeloid cells. It engages two ligands, PD-L1 (CD274) and PD-L2 (CD273). Ligand binding clusters PD-1 with the T-cell receptor and recruits the phosphatase SHP-2 via phosphorylated ITIM and ITSM motifs in the cytoplasmic tail, dampening TCR and CD28 signaling. This delivers an inhibitory checkpoint signal that limits T-cell proliferation, cytokine production, and effector function, maintaining peripheral tolerance and restraining immunopathology. Tumors and chronically infected tissues exploit this axis to drive T-cell exhaustion; blocking PD-1/PD-L1 engagement restores effector responses, the basis of checkpoint-inhibitor immunotherapy.
