Background
This is the PS/2 clone, a rat IgG2b kappa monoclonal raised against mouse CD49d (integrin alpha-4). It is one of the most widely used tools for studying alpha-4 integrin function in the mouse and is offered here in a format built for in-vivo and functional work: low endotoxin (research grade <1 EU/mg, >95% pure; ultra-low option <0.5 EU/mg, 98% pure), supplied in scalable milligram-to-gram quantities for repeat dosing across cohorts. PS/2 is a function-blocking antibody: it binds the alpha-4 subunit and interferes with alpha-4 integrin (VLA-4 and alpha-4/beta-7) engagement of its ligands, making it suited to leukocyte-adhesion and trafficking studies as well as autoimmune and inflammation models. Beyond in-vivo dosing, the clone is validated for flow cytometry, immunofluorescence, and immunohistochemistry, so the same reagent can be used to confirm target engagement and to phenotype tissues. All material is research use only (RUO) and intended for animal and in-vitro research, not therapeutic use. CD49d is integrin alpha-4 (ITGA4). It pairs with beta-1 (CD29) to form VLA-4 (alpha-4/beta-1) and with beta-7 to form the alpha-4/beta-7 integrin. VLA-4 binds VCAM-1 on activated endothelium and the CS-1 region of fibronectin, mediating firm adhesion, rolling, and transendothelial migration of lymphocytes, monocytes, eosinophils, and other leukocytes into inflamed tissue. Alpha-4/beta-7 binds MAdCAM-1 and directs lymphocyte homing to gut-associated lymphoid tissue. Because these interactions control leukocyte recruitment, alpha-4 integrin is central to inflammatory and autoimmune disease; it is the target of the clinical antibody natalizumab in humans. CD49d is expressed on most mononuclear leukocytes and is also used as an activation/memory marker on T cells. The listed UniProt accessions are mouse ITGA4 (P13612) and human ITGA4 (Q00651).
