Background
This product is an unconjugated, non-therapeutic recombinant analog of nadecnemab, supplied for research use only and built around the target GFRA3 (GDNF family receptor alpha-3). It is a human IgG4-kappa molecule that reproduces the antigen-binding specificity of the originator anti-GFRA3 antibody, allowing laboratories to interrogate GFRA3 biology and the artemin/RET signaling axis without sourcing clinical material. It is not the clinical drug and is not intended for human or veterinary use. The preparation is offered at research-grade low endotoxin (typically under 1 EU/mg, with ultra-low-endotoxin options under 0.5 EU/mg) and in bulk milligram-to-gram quantities, making it well suited to reproducible assay development and scale-up. Typical research applications include use as a binding and specificity reference standard, as an isotype-matched positive control in GFRA3 detection or neutralization workflows, as a benchmark antibody for mechanism-of-action and epitope studies, and as a tool for antibody-engineering and ADC or ADCC development pipelines. The IgG4 backbone confers minimal effector recruitment, which is useful when a predominantly ligand- or receptor-blocking readout is desired.
GFRA3 (GDNF family receptor alpha-3; UniProt O60609) is a glycosylphosphatidylinositol (GPI)-anchored cell-surface co-receptor and a member of the GDNF-family receptor alpha (GFRalpha) family. It is the preferred ligand-binding co-receptor for artemin (ARTN), one of the four GDNF-family ligands. Upon binding ARTN, GFRA3 recruits and drives autophosphorylation and activation of the RET receptor tyrosine kinase, engaging downstream cascades including RAS-MAPK (ERK), PI3K-AKT, and Src signaling. This ARTN-GFRA3-RET axis supports the development, survival, and maintenance of subsets of peripheral sensory and sympathetic neurons, and its expression is largely restricted to specific neuronal and neural-crest-derived populations. Because of its role in nociceptor sensitization and its comparatively selective tissue distribution, GFRA3 has attracted interest as a target in pain and in artemin-associated processes.
