Background
This is a research-grade biosimilar of Patecibart, supplied as an unconjugated, non-therapeutic recombinant analog built around the same target specificity as the originator antibody. Patecibart is a human IgG4-kappa monoclonal antibody directed against the endothelin receptor type A (EDNRA), a G protein-coupled receptor. Rather than the clinical product, this material is a research-use-only (RUO) reagent intended to reproduce the antigen-binding behavior of the originator so laboratories can study EDNRA engagement, run isotype- and target-matched controls, and support in-vitro and preclinical work without sourcing a therapeutic drug. Because it is a full-length human IgG4, it is useful as a benchmarking tool in binding and blocking assays, as a positive control in receptor-detection workflows, and as a reference antibody during assay development and antibody-engineering campaigns. It is offered at research grade with low endotoxin (typically <1 EU/mg, with an ultra-low <0.5 EU/mg option) and can be supplied in bulk milligram-to-gram quantities for scale-dependent studies. It is not the clinical drug and is not for human or veterinary use.
EDNRA (endothelin receptor type A; UniProt P25101) is a class A, seven-transmembrane G protein-coupled receptor and one of the two principal receptors for the endothelin peptides. It binds endothelin-1 (ET-1) with high affinity and shows selectivity for ET-1 and ET-2 over ET-3. EDNRA is expressed prominently on vascular smooth muscle cells, where ET-1 engagement couples through Gq/11 to activate phospholipase C, generating inositol trisphosphate and diacylglycerol, raising intracellular calcium, and driving sustained vasoconstriction. It also signals via Gi and G12/13 pathways to promote proliferation, migration, and hypertrophy. Through these activities EDNRA contributes to vascular tone, blood pressure regulation, and cardiac and vascular remodeling, and its signaling has been studied in pulmonary arterial hypertension, cardiovascular disease, and tumor biology. Its counterpart, EDNRB, mediates distinct and sometimes opposing effects such as ET-1 clearance and nitric-oxide-dependent vasodilation.
