Background
ICH5261 is a research-grade biosimilar of narlumosbart, supplied as an unconjugated, non-therapeutic recombinant analog of the originator anti-RANKL antibody for research use only. It is a fully human IgG4-kappa immunoglobulin engineered around the same molecular target as the clinical drug (human RANKL / TNFSF11, UniProt O14788), reproducing the antigen-binding specificity so investigators can study RANKL neutralization in controlled settings. It is not the clinical product and is not intended for human or veterinary use. Typical research applications include use as a reference or benchmarking antibody, a positive control in RANKL-binding and neutralization assays, a tool for probing the RANKL-RANK-OPG axis in vitro, and a comparator in ADCC-independent blocking studies (IgG4 has minimal effector function). Material is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg available) and in bulk milligram-to-gram quantities suitable for assay development, preclinical characterization, and in-vitro standardization. Lot-to-lot consistency supports reproducible comparisons across experiments.
TNFSF11, better known as RANKL (Receptor Activator of Nuclear factor-kappa-B Ligand; also TRANCE, OPGL), is a type II transmembrane protein of the TNF superfamily that also exists as a soluble, cleaved form. It is expressed by osteoblasts, osteocytes, activated T cells, and stromal cells. RANKL signals through its receptor RANK (TNFRSF11A) on osteoclast precursors, driving their differentiation, fusion, activation, and survival, and thereby promoting bone resorption. The decoy receptor osteoprotegerin (OPG/TNFRSF11B) competitively sequesters RANKL to restrain this pathway; the RANKL:OPG balance is a central determinant of bone remodeling. Beyond bone, RANKL contributes to lymph node organogenesis, immune regulation, and mammary gland development. Dysregulated RANKL signaling underlies osteoporosis, tumor-induced osteolysis, bone metastasis, and giant cell tumor of bone, making it a validated therapeutic and research target.
