Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-alpha-synuclein antibody exidavnemab, produced as a human IgG4-kappa molecule built around the SNCA target (UniProt P37840). It is supplied for research use only and is not the clinical drug, not for human or veterinary use. The originator is being developed to engage aggregated, pathological forms of alpha-synuclein while sparing physiological monomer, making a research-grade analog useful for probing that same aggregate-selective biology in controlled settings. Typical applications include use as a defined isotype-matched binding reagent, as a positive or reference control in immunoassays and immunohistochemistry, in neutralization or aggregate-capture experiments, in ADCC/ADC and antibody-engineering development workflows, and in preclinical model characterization. The analog is offered at research grade with low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), in in-vivo-standard formulations, and in bulk milligram-to-gram quantities to support assay development and scale-up. This description is guidance for ichorbio to review; it makes no therapeutic claims and does not reference any specific antibody clone.
SNCA encodes alpha-synuclein, a small, natively unfolded presynaptic protein highly abundant in neurons. In its physiological, often membrane-associated multimeric state it regulates synaptic vesicle trafficking, clustering, and neurotransmitter release, contributing to SNARE-complex assembly and modulating dopamine handling. Under pathological conditions the protein misfolds and self-assembles into oligomers and beta-sheet-rich amyloid fibrils that accumulate as Lewy bodies and Lewy neurites, the neuropathological hallmark of synucleinopathies including Parkinson's disease, Parkinson's disease dementia, dementia with Lewy bodies, and multiple system atrophy. Aggregated species are associated with synaptic dysfunction, mitochondrial impairment, and cell-to-cell propagation of pathology. This distinction between benign monomer and toxic aggregate underlies interest in conformation- and aggregate-selective antibodies as research tools.
