Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-CGRP antibody fremanezumab, supplied for research use only. It is a fully human IgG2-kappa immunoglobulin engineered to bind calcitonin gene-related peptide (CGRP), the same ligand targeted by the originator, making it a convenient reference reagent for laboratories studying CGRP biology and the pharmacology of anti-migraine biologics. It is not the clinical drug and is not intended for human or veterinary use. Because it reproduces the antigen-binding behaviour of the originator without a therapeutic label, it is useful as a positive control in binding and neutralisation assays, as a benchmark in developing or validating in-house anti-CGRP antibodies and ELISAs, and as a tool for probing CGRP-receptor signalling in vitro and in preclinical model systems. The material is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), in in-vivo-standard formats, and in bulk milligram-to-gram quantities suitable for assay development, scale-up, and repeated experimental use. Calcitonin gene-related peptide (CGRP) is a 37-amino-acid neuropeptide produced by alternative splicing of the calcitonin/CGRP gene (alpha-CGRP; a related beta-CGRP arises from a separate gene). It is widely distributed in central and peripheral sensory neurons, including trigeminal and dorsal root ganglia. CGRP is one of the most potent endogenous vasodilators known and acts through the CGRP receptor, a complex of the calcitonin-receptor-like receptor (CALCRL) with receptor activity-modifying protein 1 (RAMP1) plus the intracellular protein RCP, signalling largely via Gs and cyclic AMP. Beyond vasodilation, CGRP contributes to neurogenic inflammation, nociceptive signalling, and mast-cell modulation. Its release from trigeminal afferents and central sensitisation are central to migraine pathophysiology, which is why sequestering the peptide, as fremanezumab does, reduces migraine frequency.
