Background
This product is an unconjugated, non-therapeutic recombinant analog of the originator antibody evunzekibart (ATOR-1017), supplied strictly for research use only. It is a human IgG4-kappa molecule built around the extracellular domain of 4-1BB (CD137, TNFRSF9), the same costimulatory receptor engaged by the originator, and it is manufactured as a functional research reagent rather than as the clinical drug. It is not intended for human or veterinary use. The material is produced at low endotoxin (research grade <1 EU/mg, with ultra-low <0.5 EU/mg options) and is available in bulk milligram-to-gram quantities, making it well suited to work that consumes larger amounts of a defined, batch-consistent reagent. Typical research applications include use as a binding and specificity control, a benchmarking comparator for other anti-4-1BB agents, a positive control in receptor-engagement or reporter assays, a tool for studying 4-1BB agonism and Fc-gamma-receptor-dependent crosslinking, and as a starting point for conjugation, labeling, or ADCC/ADC development workflows. 4-1BB (CD137), encoded by TNFRSF9 and mapping to UniProt Q07011, is a member of the tumor necrosis factor receptor superfamily and functions as an inducible costimulatory receptor. It is expressed primarily on activated CD4+ and CD8+ T cells, natural killer cells, regulatory T cells, and some myeloid and dendritic cell populations. Its natural ligand is 4-1BBL (TNFSF9), typically presented on activated antigen-presenting cells. Engagement and clustering of 4-1BB deliver a costimulatory signal, largely through TRAF1 and TRAF2 recruitment and downstream NF-kB and MAPK activation, that enhances T-cell proliferation, cytokine production, survival, and effector and memory differentiation, and that boosts NK-cell cytotoxicity. Because agonist activity depends on receptor crosslinking, Fc-gamma-receptor engagement strongly influences signaling strength. These properties make 4-1BB a prominent target for cancer immunotherapy aimed at amplifying antitumor immune responses.
