Background
The ICH5291 Zigakibart Biosimilar (Research Grade) is an unconjugated, non-therapeutic research analog of the originator antibody zigakibart (development code BION-1301), supplied for research use only and not for clinical or diagnostic application. It is built to recapitulate the target-binding behaviour of the originator so investigators can study the biology of the pathway without sourcing clinical material. Note that the originator zigakibart is an anti-APRIL antibody (APRIL / TNFSF13), a member of the tumour necrosis factor superfamily; the target and UniProt accession recorded on this card (TNF Alpha, P01375) appear to describe a different molecule and should be corrected before publication (see flag). As a research-grade reagent it is intended for in-vitro and functional work such as ligand-binding assays, neutralisation and competition studies, and use as a positive or benchmarking control. Because the originator is directed against a human ligand and is developed for a human autoimmune kidney indication (IgA nephropathy), it is not intended as a mouse in-vivo depletion or checkpoint tool. Investigators should confirm target, format, and lot-specific specifications against their experimental needs.
The verified target of zigakibart is APRIL (A proliferation-inducing ligand; TNFSF13), a tumour necrosis factor superfamily ligand distinct from TNF-alpha. APRIL is a key survival and differentiation signal for B cells and plasma cells, acting through the receptors BCMA and TACI. In IgA nephropathy, APRIL drives class-switching and survival of plasma cells that produce galactose-deficient IgA1 (Gd-IgA1); these aberrantly glycosylated antibodies form immune complexes that deposit in the glomerular mesangium and cause kidney injury. By binding APRIL and blocking its engagement of BCMA and TACI, zigakibart reduces Gd-IgA1 production and downstream immune-complex formation. The card-listed target TNF-alpha (P01375) is a separate proinflammatory cytokine and does not reflect this drug's biology; the description here is written around APRIL, the correct target.
