Background
This is a research-grade biosimilar of sintilimab, supplied as an unconjugated, non-therapeutic recombinant analog of the originator anti-PD-1 antibody rather than the clinical drug itself. It is a fully human IgG4 monoclonal built around the programmed cell death protein 1 (PD-1) target and is intended strictly for research use only (RUO); it is not for human or veterinary use. The molecule is useful in the laboratory as a well-characterised PD-1-binding reagent for a range of applications, including target-engagement and binding studies, blockade/neutralisation assays that interrupt the PD-1/PD-L1 axis, isotype-matched positive controls in checkpoint-inhibitor screening, benchmarking of novel candidate antibodies, and as a building block for ADC or engineering workflows. It is offered at research-grade low endotoxin (<1 EU/mg), with an ultra-low endotoxin option (<0.5 EU/mg) suitable for sensitive in-vitro and preclinical standard-setting work, and is available in bulk milligram-to-gram quantities to support scale-up and assay development. Because it is produced for research rather than dosing patients, it functions as a cost-effective, consistent reference reagent for PD-1-focused programmes.
PD-1 (programmed cell death protein 1; gene PDCD1, UniProt Q15116) is an inhibitory immune-checkpoint receptor of the CD28/immunoglobulin superfamily, expressed on activated T cells and, more broadly, on B cells, NK cells, and some myeloid populations. Its cytoplasmic tail carries ITIM and ITSM motifs that, upon ligand engagement, recruit the phosphatase SHP-2 to dampen TCR and CD28 signalling, reducing proliferation, cytokine production, and effector function. PD-1 has two ligands, PD-L1 (CD274) and PD-L2 (PDCD1LG2). Under normal physiology this axis maintains peripheral tolerance and limits immunopathology, but tumours and chronically infected tissues exploit it to drive T-cell exhaustion and immune evasion. Blocking PD-1 or its ligands releases this brake and restores anti-tumour T-cell activity, the mechanistic rationale behind checkpoint-inhibitor drugs such as sintilimab.

Purification Method
This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.
Formulation
Phosphate buffered saline (PBS) pH 7.2,with no carrier protein, potassium or preservatives added. BSA and Azide free.
Storage
Stable for at least one week when stored sterile at 2-8°C. For long term storage aseptically aliquot in working volumes without diluting and store at –80°C. Avoid Repeated Freeze Thaw Cycles.
