Background
This is a research-grade, unconjugated recombinant analog of ofatumumab, a fully human IgG1 kappa antibody directed against CD20. It is manufactured as a non-therapeutic biosimilar for research use only and is not the clinical product; it is not intended for human or veterinary use. The analog reproduces the antigen-binding specificity of the originator, making it a convenient tool for laboratories that need a well-characterized anti-CD20 reagent for functional and biochemical work. Typical applications include use as a binding and neutralization reference standard, a benchmarking control against other CD20-directed molecules, a positive control in complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC) assays, and a starting scaffold for antibody-drug-conjugate or bispecific development workflows. Because it is offered at research grade with low endotoxin (research grade less than 1 EU/mg; ultra-low less than 0.5 EU/mg options) and in bulk milligram-to-gram quantities, it suits assay standardization, in-vitro potency comparisons, and preclinical model work where reproducible, well-defined material is required. Guidance provided for ichorbio to review. CD20 (encoded by MS4A1, UniProt P11836) is a non-glycosylated, four-transmembrane-domain phosphoprotein of the MS4A family expressed on the surface of B lymphocytes. It appears at the pre-B to mature B-cell stages and is lost upon differentiation into plasma cells, and it is absent from hematopoietic stem cells and most other lineages, making it a highly restricted B-cell marker. Both its amino- and carboxy-termini are cytoplasmic, and the two extracellular loops present a small epitope footprint. CD20 is not shed and is minimally internalized upon antibody binding, which favors sustained surface engagement and effector recruitment. It is thought to function in calcium flux and B-cell activation and proliferation, and it organizes within lipid rafts. These properties make CD20 a validated target for B-cell depletion, with ofatumumab binding a membrane-proximal epitope spanning both extracellular loops.
