Background
This is a research-grade, unconjugated non-therapeutic recombinant analog of nesvacumab, a fully human IgG1 monoclonal antibody directed against human Angiopoietin-2 (ANGPT2/Ang-2; UniProt O15123). It reproduces the target specificity of the originator investigational drug, which was developed as an anti-angiogenic agent (studied alone and in combination with anti-VEGF approaches in oncology and ophthalmology). Supplied for research use only, it is not the clinical product and is not intended for human or veterinary use. The format is well suited to in-vitro and preclinical laboratory work: as a positive control or reference antibody in Ang-2 binding and neutralization assays, for benchmarking biosimilar or novel anti-Ang-2 candidates, for ADC/bispecific development workflows, and for studying Ang-2 signaling in the Tie2 axis. It is available at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities to support scaled and reproducible experiments. Angiopoietin-2 (ANGPT2/Ang-2) is a secreted glycoprotein ligand of the Tie2 (TEK) receptor tyrosine kinase and a key regulator of vascular remodeling. It is stored in endothelial Weibel-Palade bodies and released rapidly upon activation, acting largely as a context-dependent antagonist of the stabilizing ligand Angiopoietin-1. By competing for Tie2, Ang-2 destabilizes quiescent, mature vessels and sensitizes the endothelium to angiogenic and inflammatory cues, priming vessels for sprouting in the presence of VEGF or for regression in its absence. Ang-2 is strongly upregulated at sites of active angiogenesis, tumor vasculature, and vascular inflammation, and its dysregulation is implicated in tumor growth, metastasis, and ocular neovascular and edematous disease. This antagonistic, destabilizing role, and the cross-talk between the Ang/Tie2 and VEGF pathways, underlies the therapeutic interest in neutralizing Ang-2.
