Background
This product is an unconjugated, non-therapeutic recombinant analog of naptumomab, supplied strictly for research use only and built around binding to the oncofetal antigen 5T4 (trophoblast glycoprotein, TPBG; UniProt Q13641). It is not the clinical agent and is not intended for human or veterinary use; the originator naptumomab estafenatox is a targeting moiety fused to a bacterial superantigen, whereas this research-grade analog is provided as a plain recombinant human IgG1 kappa without any toxin, drug, or effector payload. It is useful as a defined, reproducible reagent for interrogating 5T4 biology: as a positive binding control in flow cytometry and immunoassays, as a benchmarking or capture tool when developing and characterising other anti-5T4 antibodies, as a targeting arm in antibody-drug-conjugate or bispecific engineering workflows, and in in-vitro neutralisation, internalisation, and receptor-occupancy studies. Manufactured to research-grade specifications with low endotoxin and available in bulk milligram-to-gram quantities, it supports both in-vitro assay work and preclinical model development where a consistent, well-characterised 5T4-directed binder is required. 5T4, also called trophoblast glycoprotein (TPBG), is a heavily N-glycosylated 72 kDa type I transmembrane protein containing leucine-rich repeat domains in its extracellular region. It is termed an oncofetal antigen because expression is high in placental trophoblast and early development but tightly restricted in normal adult tissues, while it is re-expressed on the surface of many carcinomas, including colorectal, ovarian, gastric, renal, breast, and non-small-cell lung cancers. This tumour-selective distribution, combined with cell-surface accessibility, makes 5T4 an attractive target for antibody-based therapeutics, ADCs, and immunotherapies. Functionally, 5T4 is associated with epithelial-to-mesenchymal transition, altered cell adhesion and motility, and modulation of Wnt and CXCL12/CXCR4 signalling, and its expression frequently correlates with more advanced disease and poorer prognosis, supporting its use both as a tumour marker and as a targeting antigen.
