Background
This product is a research-grade, unconjugated recombinant analog of labetuzumab, an anti-CEACAM5 (CEA / CD66e) humanized IgG1 kappa antibody. It is supplied strictly for research use only and is not the clinical therapeutic; it is not intended for human or veterinary use. As a non-therapeutic biosimilar built around the CEACAM5 target, it reproduces the antigen-binding specificity of the originator molecule in a format suited to laboratory work. Typical applications include use as a defined, lot-consistent binding reagent, as a positive control or benchmark antibody in assay development, and as a building block for antibody-drug conjugate (ADC) and immunoconjugate development programs where a well-characterized anti-CEA arm is needed. It is offered as a full-length human IgG1 kappa molecule, enabling Fc-dependent readouts such as ADCC in vitro. Material is available at low-endotoxin research grade (<1 EU/mg), with ultra-low-endotoxin (<0.5 EU/mg) and bulk milligram-to-gram quantities suitable for preclinical and in-vitro standards, screening, and reagent-scale workflows. This guidance is provided for ichorbio to review.
CEACAM5 (carcinoembryonic antigen-related cell adhesion molecule 5), historically known as CEA and clustered as CD66e, is a heavily glycosylated, GPI-anchored member of the immunoglobulin superfamily and the CEA family. Its extracellular region comprises seven immunoglobulin-like domains: an N-terminal IgV-like domain followed by six IgC2-like domains. CEACAM5 mediates homotypic and heterotypic cell-cell adhesion and modulates cell fate through effects on differentiation, apoptosis (including anoikis resistance), and cell polarity. It is normally expressed on the apical surface of gastrointestinal epithelium but is markedly overexpressed and often mislocalized in many epithelial carcinomas, including colorectal, gastric, pancreatic, and lung adenocarcinomas. Shed soluble CEA in serum is a widely used tumor biomarker. This tumor-associated, surface-accessible expression pattern underlies CEACAM5's use as a target for antibody-based imaging, radioimmunotherapy, and ADC approaches.
