This product is an unconjugated, non-therapeutic recombinant analog of nivolumab, a fully human IgG4 kappa antibody directed against programmed cell death protein 1 (PD-1, PDCD1; UniProt Q15116). It is supplied for research use only and is not the clinical drug, nor intended for human or veterinary use. Built around the PD-1 target, it is useful as a defined, reproducible reagent for studies of the PD-1/PD-L1 checkpoint axis: as a positive control or benchmarking tool in binding and blocking assays, for characterizing PD-1 expression on T cells, and as a comparator in biosimilarity, ADCC/ADC-development, and assay-qualification work. The IgG4 backbone limits effector function, consistent with a checkpoint-blocking mechanism rather than target-cell depletion. It is manufactured at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), available in bulk milligram-to-gram quantities, and formatted as an in-vivo-standard-grade reagent to support consistent preclinical and in-vitro experimentation. PD-1 (programmed cell death protein 1, encoded by PDCD1; UniProt Q15116) is a type I transmembrane immunoinhibitory receptor of the CD28/B7 family, expressed on activated T cells, B cells, NK cells, and myeloid subsets. Its cytoplasmic tail carries an ITIM and an ITSM that recruit the phosphatase SHP-2 upon ligand engagement. Binding of its ligands PD-L1 (CD274) and PD-L2 (CD273) dampens TCR and CD28 signaling, attenuating T-cell proliferation, cytokine production, and survival. This pathway maintains peripheral tolerance and limits immunopathology, but is co-opted in the tumor microenvironment and chronic infection to drive T-cell exhaustion. Blocking PD-1/PD-L1 engagement restores effector T-cell activity, the rationale underlying checkpoint-inhibitor immunotherapy.

This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

Phosphate buffered saline (PBS) pH 7.2,with no carrier protein, potassium or preservatives added. BSA and Azide free.
Storage
Stable for at least one week when stored sterile at 2-8°C. For long term storage aseptically aliquot in working volumes without diluting and store at –80°C. Avoid Repeated Freeze Thaw Cycles.
