Background
ICH5130 is an unconjugated, non-therapeutic recombinant analog of anifrolumab, engineered around the same target, interferon-alpha/beta receptor subunit 1 (IFNAR1). It is supplied for research use only (RUO) and is not the clinical drug; it is not intended for human or veterinary use. Like the originator, it is a human IgG1-kappa built with the triple Fc mutation (L234F/L235E/P331S numbering here annotated L351F/L352E/P448S) that markedly reduces FcgammaR and complement engagement, so activity is driven by receptor binding rather than effector function. As an IFNAR1-directed binder it is useful as a positive control or benchmark reagent in receptor-blockade and type I interferon signaling assays, for antagonist-antibody characterization, epitope and competition studies, and as a tool in in-vitro and preclinical models of interferon biology. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg option) and in bulk milligram-to-gram quantities to support screening, assay development, and in-vivo standard workflows. IFNAR1 (UniProt P17181) is one of two subunits of the heterodimeric type I interferon receptor; together with IFNAR2 it forms the signaling complex for all type I interferons, including the multiple IFN-alpha subtypes, IFN-beta, IFN-omega, IFN-epsilon and IFN-kappa. Ligand engagement brings IFNAR1 and IFNAR2 together, activating the receptor-associated Janus kinases TYK2 (on IFNAR1) and JAK1 (on IFNAR2). This drives phosphorylation of STAT1 and STAT2, assembly of the ISGF3 complex with IRF9, and transcription of interferon-stimulated genes. Type I IFN signaling shapes antiviral defense, innate and adaptive immunity, and a self-amplifying feedback loop. Sustained IFNAR1 signaling and an elevated interferon gene signature are implicated in autoimmune conditions such as systemic lupus erythematosus, making IFNAR1 a well-validated blockade target.
