Background
This is a research-grade, unconjugated recombinant analog of cemiplimab, an anti-PD-1 antibody, supplied strictly for research use only and not as the clinical therapeutic. It is built as a human IgG4 (S228P-stabilized) kappa antibody directed against human PD-1 (PDCD1, UniProt Q15116), reproducing the target specificity of the originator so investigators can probe PD-1 biology and checkpoint signaling in controlled settings. Typical applications include binding and blocking assays, use as a positive control or reference antibody in immune-checkpoint studies, benchmarking of biosimilar or novel anti-PD-1 candidates, and as a tool reagent in ADCC/engineering and antibody-development workflows. It is offered at low endotoxin levels (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and available in bulk milligram-to-gram quantities to support in-vitro assays, preclinical model work, and scale-dependent screening. Because it is a non-therapeutic analog, it is intended only for laboratory investigation and is not for human or veterinary use. It lets labs study PD-1 blockade without sourcing clinical drug product.
PD-1 (programmed cell death protein 1; gene PDCD1, UniProt Q15116) is an inhibitory immune checkpoint receptor of the CD28/B7 family expressed on activated T cells, B cells, NK cells, and other immune subsets. It carries an immunoreceptor tyrosine-based inhibitory motif (ITIM) and switch motif (ITSM) in its cytoplasmic tail; engagement by its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2) recruits the phosphatase SHP-2, dampening TCR and CD28 signaling to attenuate T-cell activation, proliferation, and cytokine production. This pathway maintains peripheral tolerance and limits immunopathology, but tumors and chronic infections co-opt it to drive T-cell exhaustion and immune escape. Antibodies that block the PD-1/PD-L1 axis restore effector T-cell function, the basis of checkpoint-inhibitor immunotherapy.
