Background
This product is an unconjugated, non-therapeutic recombinant analog of TSR-033, an anti-human LAG3 (Lymphocyte Activation Gene 3) antibody, supplied strictly for research use only. It is not the clinical drug and is not intended for human or veterinary use. The originator is a human IgG4 (S228P-stabilized) antibody that binds LAG3 and blocks its engagement with MHC class II, relieving LAG3-mediated inhibition of T cells; the IgG4 backbone is chosen to minimize effector functions such as ADCC and CDC. This research-grade analog reproduces the target specificity and the human IgG4/kappa framework, making it useful as a defined, reproducible tool for LAG3 pathway studies. Typical research applications include use as a binding and blocking reference standard, a positive control in checkpoint assays, a benchmarking reagent against other anti-LAG3 molecules, and a building block for ADC or engineering workflows. It is manufactured with low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) and is available in bulk milligram-to-gram quantities to support in-vitro and preclinical work.
LAG3 (Lymphocyte Activation Gene 3; CD223; UniProt P18627) is an inhibitory immune checkpoint receptor structurally related to CD4, expressed on activated CD4+ and CD8+ T cells, regulatory T cells, NK cells, NKT cells, B cells and plasmacytoid dendritic cells. Its canonical ligand is MHC class II, which it binds with higher affinity than CD4; additional functional ligands include FGL1, and it also associates with the TCR-CD3 complex, galectin-3 and LSECtin. Engagement delivers negative signals that limit T-cell proliferation, cytokine production and activation, and LAG3 is strongly upregulated on exhausted T cells within the tumor microenvironment. Because it acts through pathways largely non-redundant with PD-1, LAG3 is a leading target for combination cancer immunotherapy, and blocking LAG3 can restore effector T-cell function.
