RMP1-14 is a rat IgG2a, kappa monoclonal antibody raised against mouse PD-1 (Pdcd1, CD279; UniProt Q02242). It is one of the most widely adopted clones for functional interrogation of the PD-1 pathway in mouse models, recognizing the extracellular domain of mouse PD-1 and blocking its engagement by the ligands PD-L1 (CD274) and PD-L2 (CD273). The rat IgG2a backbone is a common choice for in-vivo mouse work, and RMP1-14 is routinely used for checkpoint-blockade studies, alone or combined with other immunomodulatory or targeted agents. This preparation is supplied at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities, making it suited to in-vivo dosing, ex-vivo functional assays, and larger experimental cohorts. Validated applications include blocking, flow cytometry, functional assays, and Western blot. For research use only (RUO); not for diagnostic or therapeutic use. As with any lot and model, users should validate performance under their own experimental conditions. PD-1 (programmed cell death protein 1; CD279) is an inhibitory immune-checkpoint receptor of the CD28/CTLA-4 immunoglobulin superfamily, expressed on activated T cells and, to varying degrees, B cells, NK cells, and myeloid subsets. Its expression is induced upon antigen-driven lymphocyte activation. Engagement by its ligands PD-L1 (CD274) and PD-L2 (CD273) recruits the phosphatase SHP-2 (PTPN11) to the receptor's cytoplasmic ITIM/ITSM motifs, dampening proximal TCR and CD28 signaling and thereby limiting T-cell activation, proliferation, and cytokine production. This axis is essential for peripheral tolerance and restraining immunopathology, but tumors and chronic infections co-opt it to drive T-cell exhaustion and immune evasion. Blockade of PD-1 or its ligands can restore effector T-cell function, the rationale underpinning checkpoint-blockade immunotherapy.

Application Notes
Blocking: Clone RMP1-14 antibody has been shown to block the binding of both mouse PD-L1-Ig and mouse PD-L2-Ig to PD-1. For immune checkpoint antibodies 200 ug per mouse per injection is the standard dosage in the literature. For this anti-PD-1 (clone RMP1-14), we suggest to inject 200 ug/mouse ip generally on days 3, 6 and 9, post injection of tumor. Sometime up to six injections depending on the length of the experiment may be required. Each investigator should determine their own optimal working dilution for specific applications.
