Background
This is bulk anti-human CD3 clone UCHT-1, a mouse IgG1 monoclonal raised against human CD3 (UniProt P07766, the CD3-epsilon subunit). UCHT-1 is one of the most widely used and best-characterised anti-human CD3 clones, recognising the epsilon chain of the TCR/CD3 complex on mature human T cells. It is validated across a broad application range, including flow cytometry, immunoprecipitation, western blot, frozen-section IHC, CyTOF/mass cytometry, and functional T-cell assays. Supplied in low-endotoxin, bulk (mg to gram) format for research use only, it is well suited to functional and high-throughput work where endotoxin, aggregation, and lot consistency matter, such as plate-bound or soluble T-cell activation, ex vivo expansion protocols, and immune-monitoring panels. Because the target is human and this is a human-reactive research reagent, it is intended for in-vitro, ex-vivo, and human-sample applications rather than mouse in-vivo dosing models. The bulk, low-endotoxin presentation makes it economical for large screens and repeated functional assays. CD3 is the invariant signalling module of the T-cell receptor (TCR) complex, composed of the CD3-gamma, CD3-delta, and CD3-epsilon chains together with a zeta-chain homodimer, assembled non-covalently around the antigen-binding TCR-alpha/beta (or gamma/delta) heterodimer. Clone UCHT-1 binds the CD3-epsilon subunit encoded near UniProt P07766. While the TCR provides antigen specificity, it lacks intrinsic signalling capacity; the CD3 chains carry the immunoreceptor tyrosine-based activation motifs (ITAMs) that, upon TCR engagement, are phosphorylated by Lck and recruit ZAP-70 to initiate downstream activation, calcium flux, and proliferation. CD3 is expressed exclusively on T lineage cells, making it the definitive pan-T-cell marker in immunophenotyping. Antibodies to CD3-epsilon such as UCHT-1 can crosslink the complex to trigger TCR-independent T-cell activation, which underlies their use in functional stimulation assays.

Application Notes
Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing CD3. A suggested method would be to stain cells expressing CD3 with 2.0 µg per million cells in a total staining volume of 100 µl followed by Goat Anti-Mouse IgG (H&L)-R-phycoerythrin. Each investigator should determine their own optimal working dilution for specific applications.
