Background
This product is a research-grade biosimilar of crovalimab: an unconjugated, non-therapeutic monoclonal analog built around the same target as the originator drug and supplied for research use only. It is not a clinical drug and is not intended for therapeutic or diagnostic use. Crovalimab is an anti-complement C5 antibody engineered with sequential monoclonal antibody recycling technology (SMART), which combines pH-dependent (recycling) antigen binding, isoelectric point engineering, and neonatal Fc receptor (FcRn) engineering. These features drive efficient uptake and endosomal disposal of bound C5 and FcRn-mediated recycling of the antibody, enabling infrequent, low-volume subcutaneous dosing in the therapeutic setting for paroxysmal nocturnal hemoglobinuria (PNH). Notably, crovalimab binds the C5 beta-chain, distinguishing it from alpha-chain-directed C5 inhibitors such as eculizumab and ravulizumab. The biosimilar format is designed for functional and comparative in-vitro work: complement-pathway neutralisation assays, target-binding and epitope characterisation, and use as a reference or control molecule. The listed isotype is human IgG1/kappa and the annotated target is human C5 (UniProt P01031). Investigators should confirm lot-specific specifications, formulation, and endotoxin level for their intended application. Complement component 5 (C5; UniProt P01031) is a central effector protein of the complement cascade. C5 is cleaved by C5 convertases into the anaphylatoxin C5a, a potent pro-inflammatory chemoattractant, and C5b, which nucleates assembly of the terminal complement complex (C5b-9, the membrane attack complex) that lyses target cells. Uncontrolled terminal complement activation drives intravascular hemolysis and thrombosis in disorders such as PNH and contributes to atypical hemolytic uremic syndrome and other complement-mediated conditions. Therapeutic and research anti-C5 antibodies act by blocking C5 cleavage, thereby suppressing both C5a generation and downstream MAC formation. Crovalimab engages an antigenic determinant on the C5 beta-chain and additionally can limit C5b6 deposition on membranes, adding a distinct mode of terminal-pathway inhibition compared with alpha-chain binders. C5 is highly abundant in plasma, so binding stoichiometry and target turnover are important considerations in complement assays.
