Background
ICH5250 is a research-grade, unconjugated recombinant analog of the anti-TGF-beta2 antibody Lerdelimumab, built as a human IgG4/lambda molecule that binds and neutralizes human transforming growth factor beta-2 (TGFB2, UniProt P61812). It reproduces the target specificity of the originator drug but is manufactured strictly for research use only; it is not the clinical product and is not intended for human or veterinary use. The format is useful for laboratory work where a well-characterized TGF-beta2-neutralizing reagent is needed: as a positive control or benchmark in binding and neutralization assays, as a tool for probing TGF-beta2-driven signaling in vitro, and as a scaffold for antibody-engineering and biosimilar-comparability studies. Because the antibody is supplied unconjugated, it can be paired with secondary detection systems or used directly in functional readouts. Preparations are offered at research grade with low endotoxin (typically <1 EU/mg, with an ultra-low <0.5 EU/mg option) and are available in bulk milligram-to-gram quantities to support assay development, screening, and preclinical model work requiring consistent lot-to-lot performance. TGF-beta2 is one of three mammalian TGF-beta isoforms, secreted as a latent complex and activated to a disulfide-linked homodimer that signals through the type II (TGFBR2) and type I (TGFBR1/ALK5) serine/threonine kinase receptors, driving SMAD2/3 phosphorylation and transcriptional programs. TGF-beta2 regulates cell proliferation, differentiation, adhesion, migration, and extracellular matrix deposition, and exerts potent immunosuppressive effects, including suppression of IL-2-dependent T-cell growth. It has prominent roles in ocular and cardiac development, wound healing, and fibrosis, and elevated TGF-beta2 in the tumor microenvironment is associated with immune evasion and poor prognosis. Unlike the widely studied TGF-beta1, TGF-beta2 shows lower affinity for TGFBR2 alone and often depends on the co-receptor betaglycan (TGFBR3) for efficient signaling, which contributes to its distinct, context-dependent biology.
