Background
This antibody targets mouse CD40 using clone FGK4.5, a rat IgG2a monoclonal raised against the mouse antigen. FGK4.5 is one of the most widely used agonistic anti-CD40 clones in immunology, valued for its ability to engage and activate CD40 signalling on antigen-presenting cells rather than simply mark the target. That functional agonist behaviour makes it well suited to in-vivo and mechanistic studies of dendritic cell and macrophage licensing, T-cell priming, and antitumour immunity, as well as to flow cytometry and western blot applications listed for the product. The preparation is manufactured for in-vivo work: low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg), supplied in bulk milligram-to-gram quantities, and provided as a research-use-only (RUO) reagent. The rat IgG2a format allows use with matched rat IgG2a isotype controls. Low endotoxin and bulk formatting reduce confounding innate immune activation and support the repeated dosing and larger cohorts typical of syngeneic tumour and immunisation models. CD40 (UniProt P27512) is a type I transmembrane glycoprotein of the tumour necrosis factor receptor (TNFR) superfamily. It is expressed on antigen-presenting cells including dendritic cells, macrophages, and B cells, as well as on some endothelial and epithelial cells. Its natural ligand is CD40L (CD154), expressed largely on activated CD4+ T cells and platelets. CD40-CD40L engagement is a central costimulatory axis: on dendritic cells it drives maturation, upregulation of MHC and costimulatory molecules, and cytokine production, thereby "licensing" APCs to prime CD8+ T-cell responses. On B cells it promotes proliferation, germinal centre formation, immunoglobulin class switching, and affinity maturation. Because CD40 stimulation can convert tolerogenic APCs into activating ones, agonism of this pathway is a major strategy for boosting antitumour and antiviral immunity.
