This is the widely used clone 22D1, an Armenian hamster IgG1 kappa monoclonal that recognizes mouse CD209b (SIGN-R1), a marginal-zone macrophage C-type lectin. It is offered as a low-endotoxin, in-vivo-ready preparation in bulk milligram-to-gram quantities for research use only, making it well suited to functional and in-vivo studies in mice where endotoxin contamination and formulation volume are limiting. Because 22D1 binds the carbohydrate-recognition region of SIGN-R1, it is frequently applied not only for detection (flow cytometry, frozen-section IHC, immunofluorescence, immunocytochemistry, Western blot, ELISA) but also as a blocking/functional reagent to interrupt SIGN-R1-mediated ligand capture in vivo and ex vivo. The hamster host and cross-reactivity profile make it a standard tool for labelling and functionally probing splenic marginal-zone and lymph-node medullary macrophage populations. CD209b, also called SIGN-R1, is a type II transmembrane C-type lectin and one of the five mouse homologues of human DC-SIGN (CD209). It is expressed at high levels on splenic marginal-zone macrophages and lymph-node medullary macrophages. Through its calcium-dependent carbohydrate-recognition domain it binds high-mannose oligosaccharides and dextran, and it captures bacterial capsular polysaccharides such as those of Streptococcus pneumoniae, contributing to clearance of encapsulated organisms and to T-independent antibody responses. SIGN-R1 also participates in innate recognition and complement activation: it can bind C1q and promote classical complement C3 deposition on polysaccharide, and it has been reported to associate with pattern-recognition partners on the macrophage surface. Its restricted, well-defined expression makes it a valuable marker of marginal-zone macrophages.
