Background
ICH5462 is a research-grade biosimilar of dalnicastobart, supplied as an unconjugated, non-therapeutic recombinant analog for research use only. It is built around the sequence and binding characteristics of the originator antibody, a human IgG1 (kappa light chain) directed against human CD40 (TNFRSF5, UniProt P25942). Dalnicastobart (also reported as LVGN-7409) is an investigational agonistic anti-CD40 antibody explored in oncology, where CD40 engagement is intended to activate antigen-presenting cells and support anti-tumor immunity. This product is not a formulated drug and is not intended for clinical or therapeutic use; it is a laboratory reagent for in-vitro and functional characterization work. Because CD40 is a human target, this molecule is primarily suited to studies of human CD40 biology rather than routine mouse in-vivo dosing models. It is manufactured to low-endotoxin research standards and is available in bulk milligram-to-gram quantities, making it appropriate for assay development, comparability studies, and mechanistic experiments requiring a defined, sequence-matched anti-CD40 IgG1 reference reagent.
CD40 (TNFRSF5, UniProt P25942) is a type I transmembrane receptor of the tumor necrosis factor receptor superfamily, expressed on dendritic cells, B cells, macrophages, and other antigen-presenting cells, as well as on some non-immune and tumor cells. Its natural ligand is CD40L (CD154), expressed largely on activated CD4+ T cells. CD40-CD40L engagement is a central co-stimulatory axis: it licenses dendritic cells to prime CD8+ effector T-cell responses and drives B-cell activation, proliferation, class-switch recombination, and differentiation. Signaling proceeds through TRAF adaptor recruitment and downstream NF-kB and MAPK pathways. Because of this immunostimulatory role, agonistic CD40 targeting is an active area of cancer immunotherapy, while CD40 blockade is studied in autoimmune and transplant settings. CD40 is a human-restricted target here, so this reagent is most relevant to human CD40 pathway studies.
