Background
ICH5323 is a research-grade biosimilar of abiprubart (also known as KPL-404), supplied as an unconjugated, non-therapeutic analog built around the CD40 target. It is a humanized IgG4-kappa antibody matching the originator's format, in which the IgG4 backbone provides a functionally attenuated Fc so that binding blocks CD40 signaling without driving effector-mediated cell killing. The originator abiprubart is an investigational immunomodulator being developed to interrupt the CD40-CD154 (CD40 ligand) costimulatory axis, and has been evaluated in autoimmune indications including rheumatoid arthritis and Sjogren's disease. This product is intended for research use only and is not for human or clinical use. It is manufactured to in-vivo research standards with low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), and is available in bulk milligram-to-gram quantities suitable for larger studies. It is a useful reference reagent for assay development, binding and blocking studies, and as a benchmark control when characterizing anti-CD40 candidates targeting the same costimulatory pathway. CD40 (UniProt P25942) is a type I transmembrane receptor of the TNF receptor superfamily, expressed on B cells, dendritic cells, macrophages, and other antigen-presenting cells, as well as some non-immune cells. Its ligand, CD40L (CD154), is expressed mainly on activated CD4+ T cells. Engagement of CD40 by CD154 delivers a central costimulatory signal that drives B-cell activation, germinal center formation, immunoglobulin class switching, and affinity maturation, and licenses dendritic cells to prime T-cell responses. Because this axis is pivotal to humoral and Type 1 immunity, blocking CD40-CD154 interaction is an attractive strategy for suppressing pathogenic autoantibody production and T-cell-dependent inflammation in autoimmune disease, while a non-depleting, effector-silent format aims to modulate signaling without destroying the CD40-expressing cells.
