Background
This product is an unconjugated, non-therapeutic recombinant analog of lucatumumab, a fully human anti-CD40 antagonist antibody, supplied for research use only. It is built around the human CD40 target and formatted as a human IgG1(kappa) with an S131A framework substitution, providing a reproducible tool for interrogating CD40-CD40L (CD154) signaling in vitro and in preclinical models. Typical research applications include use as a binding and blocking reference standard, a positive control in CD40-directed assay development, a benchmarking reagent for comparing new anti-CD40 candidates, and a building block for ADCC or antibody-drug-conjugate feasibility work. It is not the clinical drug and is not intended for human or veterinary use. Material is offered at research-grade low endotoxin (<1 EU/mg), with an ultra-low-endotoxin option (<0.5 EU/mg) suitable for endotoxin-sensitive cell and in-vivo standard workflows, and is available in bulk milligram-to-gram quantities to support scale-up, screening campaigns, and longitudinal studies requiring lot consistency. CD40 (TNFRSF5; UniProt P25942) is a type I transmembrane costimulatory receptor of the TNF receptor superfamily, expressed on B cells, dendritic cells, macrophages, and many non-hematopoietic and tumor cells. Its ligand, CD40L (CD154), is expressed largely on activated CD4+ T cells and platelets. CD40 engagement recruits TRAF adaptors and activates NF-kappaB, MAPK, and PI3K pathways, driving B-cell proliferation, immunoglobulin class-switch recombination, germinal center formation, and antigen-presenting-cell maturation with upregulated costimulatory molecules and cytokines. This axis is central to humoral immunity and T-cell priming. Aberrant CD40 signaling contributes to autoimmunity, inflammation, and the survival of CD40-expressing malignancies, making CD40 a target for both antagonist (signal-blocking) and agonist (immune-activating) antibody strategies.
