Background
This product is a research-grade, unconjugated recombinant analog of mitazalimab, an agonistic antibody directed against human CD40. It is a non-therapeutic biosimilar intended strictly for research use, not for human or veterinary application. Supplied unconjugated as an intact human IgG1 (lambda light chain) format, it reproduces the CD40-directed binding specificity of the originator molecule and is useful as a well-defined tool for probing CD40 biology and CD40-agonist pharmacology in the laboratory. Typical applications include use as a binding and functional reference standard, a positive or benchmarking control in receptor-engagement and agonism assays, an isotype-matched comparator in antibody-characterization workflows, and a reagent for in-vitro assay development. Because it is offered at research grade, it is available in bulk milligram-to-gram quantities and in low-endotoxin preparations (research grade <1 EU/mg; ultra-low <0.5 EU/mg) suitable for sensitive cell-based and preclinical model work. It carries no therapeutic formulation claims and is provided for functional and biochemical characterization only.
CD40 (TNFRSF5; UniProt P25942) is a type I transmembrane costimulatory receptor of the TNF receptor superfamily. It is expressed on antigen-presenting cells including dendritic cells, B lymphocytes, macrophages and monocytes, and also on some endothelial and epithelial cells and various tumors. Its natural ligand is CD40L (CD154), expressed largely on activated CD4+ T cells and platelets. Ligation triggers receptor clustering and recruitment of TRAF adaptor proteins, activating NF-kB, MAPK and other pathways. In B cells this drives proliferation, germinal-center formation, immunoglobulin class-switch recombination and affinity maturation; in dendritic cells and macrophages it promotes maturation, upregulation of costimulatory molecules, and pro-inflammatory cytokine secretion. Because CD40 engagement can license antigen-presenting cells and prime cytotoxic T-cell responses, agonistic CD40 antibodies are studied as immune-activating agents in oncology, contrasting with CD40/CD40L-blocking approaches explored in autoimmunity and transplantation.
