Background
This is an unconjugated, non-therapeutic recombinant analog of the originator antibody sotigalimab, an agonistic anti-CD40 monoclonal built around the human CD40 target (UniProt P25942). It is supplied for research use only and is not the clinical drug, nor intended for human or veterinary use. The molecule is a human IgG1-kappa carrying engineered Fc substitutions (S267E/E356D/M358L) that bias Fc-gamma receptor engagement to enhance agonistic crosslinking, plus a C-terminal lysine deletion (KDel) for charge homogeneity. As a research reagent it is useful as a binding and CD40-agonism positive control, for target-engagement and epitope-competition assays, for comparing Fc-engineered versus wild-type agonism, for ADCC/effector-function and FcγR-dependence studies, and as a reference standard in bioassay and biosimilarity development. It is offered at research grade with low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options), as an in-vivo-grade standard where applicable, and in bulk milligram-to-gram quantities for screening and preclinical workflows. No clone information is implied. CD40 (UniProt P25942; TNFRSF5) is a type I transmembrane receptor of the TNF receptor superfamily, expressed on antigen-presenting cells including B cells, dendritic cells, macrophages and monocytes, as well as on many non-immune and tumor cells. Its natural ligand is CD40L (CD154), a TNF-family trimer expressed largely by activated CD4+ T cells. CD40 ligation trimerizes the receptor and recruits TRAF adaptor proteins, activating NF-kB, MAPK and PI3K signaling. This drives B-cell activation, proliferation, germinal-center formation, immunoglobulin class-switch recombination and affinity maturation, and licenses dendritic cells to prime cytotoxic T cells. Because CD40 agonism can activate APCs and promote antitumor immunity, agonistic anti-CD40 antibodies are studied in cancer immunotherapy, where Fc-gamma-receptor-dependent crosslinking is often required for full agonistic activity.
