Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-TLR4 antibody paridiprubart, supplied for research use only. It reproduces the target specificity of the originator molecule, binding human Toll-like receptor 4 (TLR4, UniProt O00206), and is intended as a laboratory reagent rather than a clinical drug. It is not for human or veterinary use. As a research-grade biosimilar it is well suited to functional and mechanistic studies: as a positive or benchmarking control alongside other TLR4-directed reagents, for in-vitro receptor-binding and neutralisation assays, for probing TLR4-dependent signalling in cell-based systems, and as a starting reagent for ADCC or antibody-drug-conjugate development workflows. The human IgG1/kappa framework supports Fc-mediated assay formats and epitope-competition studies. Material is offered at low endotoxin (research grade <1 EU/mg; ultra-low <0.5 EU/mg) and in bulk milligram-to-gram quantities, making it appropriate for reproducible in-vitro work and preclinical model characterisation where consistent, well-defined anti-TLR4 reagent is required. Toll-like receptor 4 (TLR4) is a type I transmembrane pattern-recognition receptor and a central sensor of innate immunity. Its best-characterised ligand is lipopolysaccharide (LPS) from Gram-negative bacteria, which it recognises together with the accessory proteins MD-2 (LY96), CD14 and LBP. Ligand-induced dimerisation of the TLR4/MD-2 complex triggers intracellular signalling through the TIR-domain adaptors MyD88 and TRIF, activating NF-kB and IRF3 pathways and driving production of pro-inflammatory cytokines and type I interferons. Beyond LPS, TLR4 responds to endogenous damage-associated molecular patterns (DAMPs) such as HMGB1, S100 proteins and heat-shock proteins, linking it to sterile inflammation and tissue injury. Because of this role, TLR4 is studied extensively in sepsis, acute lung injury, ischemia-reperfusion, autoimmunity and inflammatory disease, and is a recognised therapeutic target for modulating excessive innate immune activation.
