Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-CD40 agonist antibody giloralimab (also known by its development code ABBV-927), supplied strictly for research use only. It is a human IgG1, kappa monoclonal built around the human CD40 (TNFRSF5) target and is not the clinical drug; it is not intended for human or veterinary use. As a research-grade biosimilar, it is manufactured to be functionally comparable to the originator so investigators can use it as a well-defined reagent for studying CD40 engagement and agonistic signaling. Typical research applications include use as a positive control or benchmark in CD40 binding and immune-activation assays, in-vitro dendritic cell, B cell and monocyte activation studies, ADCC/effector-function characterization, and antibody-development or comparability workflows. The material is offered at low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg) and in bulk milligram-to-gram quantities, making it suitable for larger in-vitro screens, assay standardization, and preclinical model work where lot-to-lot consistency and low endotoxin are important.
CD40 (TNFRSF5, UniProt P25942) is a type I transmembrane costimulatory receptor of the tumor necrosis factor receptor superfamily. It is expressed on antigen-presenting cells including dendritic cells, B cells, macrophages and monocytes, as well as on some endothelial and epithelial cells and a range of tumors. Its natural ligand is CD40L (CD154), a trimeric molecule expressed predominantly on activated CD4+ T helper cells. Ligand engagement clusters CD40 and recruits TRAF adaptor proteins, driving NF-kB, MAPK and other signaling pathways. On antigen-presenting cells this upregulates costimulatory molecules (CD80/CD86), promotes cytokine release, and enhances antigen presentation, thereby licensing APCs and supporting T cell priming. In B cells, CD40 signaling is central to germinal-center formation, immunoglobulin class switching and affinity maturation. Because CD40 links innate and adaptive immunity, agonistic engagement is of broad interest in immuno-oncology research.
