Background
This product is a research-grade, unconjugated recombinant analog of iscalimab, an anti-CD40 antibody, supplied strictly for research use only and not for human or veterinary use. It is a human IgG1-kappa built around the CD40 target (UniProt P25942) to reproduce the binding specificity of the originator molecule while being manufactured to research specifications rather than as a therapeutic. The format is a non-therapeutic biosimilar intended as a functional tool: a defined, reproducible reagent for studying CD40 engagement and blockade in vitro and in preclinical settings. It is useful as a positive or isotype-matched control in binding and blocking assays, as a benchmarking standard when characterising new anti-CD40 candidates, and as a starting reagent for ADCC or ADC development workflows. Because iscalimab is engineered as a non-depleting, pathway-blocking antibody, the analog is well suited to experiments probing CD40-CD40L signalling interruption. It is available at low endotoxin (research grade typically under 1 EU/mg, with ultra-low options under 0.5 EU/mg) and in bulk milligram-to-gram quantities to support scale-up and repeated experiments.
CD40 (UniProt P25942) is a type I transmembrane glycoprotein of the tumour necrosis factor receptor superfamily (TNFRSF5). It is expressed on antigen-presenting cells including B cells, dendritic cells, macrophages and monocytes, as well as on endothelial and epithelial cells and many tumours. Its ligand is CD40L (CD154), presented largely by activated CD4-positive T cells. CD40-CD40L engagement drives trimerisation of the receptor and recruitment of TRAF adaptor proteins, activating NF-kappaB, MAPK and other pathways. Functionally this delivers key costimulation: B-cell activation, germinal-centre formation, immunoglobulin class switching and affinity maturation, dendritic-cell licensing, and macrophage activation. Because the axis is central to T-cell-dependent humoral immunity and to sustaining inflammatory and autoimmune responses, CD40 is a prominent target for pathway-blocking approaches in transplantation and autoimmunity, which is the biology iscalimab is designed to interrupt.
