Its natural ligand is CD40L (CD154), presented mainly by activated CD4+ T cells. Engagement of CD40 drives downstream signaling through TRAF adaptors and the NF-kB pathway, promoting APC maturation, upregulation of co-stimulatory molecules, cytokine production, B-cell activation and class switching, and priming of T-cell responses. Because CD40 ligation can license dendritic cells and enhance anti-tumor immunity, agonistic anti-CD40 antibodies are of substantial interest in immuno-oncology. The originator of this biosimilar is an agonist designed to cluster and activate CD40, with Fc engineering intended to enhance effector engagement. UniProt P25942 corresponds to human CD40, consistent with the card annotation.
