Background
This product is a research-grade biosimilar of cifurtilimab, supplied as an unconjugated, non-therapeutic analog of the originator antibody for research use only. It is not a clinical drug. The molecule is a human IgG1, kappa antibody built around the CD40 (TNFRSF5) target that the originator engages, allowing investigators to model the binding and functional behavior of the reference agent in controlled laboratory settings. Cifurtilimab (also known in the literature as SEA-CD40) is described as an agonistic, effector-enhanced anti-CD40 antibody, and this analog is intended to recapitulate target engagement rather than to serve as an approved therapeutic. Engagement of CD40 drives downstream signaling through TRAF adaptors and the NF-kB pathway, promoting APC maturation, upregulation of co-stimulatory molecules, cytokine production, B-cell activation and class switching, and priming of T-cell responses. Because CD40 ligation can license dendritic cells and enhance anti-tumor immunity, agonistic anti-CD40 antibodies are of substantial interest in immuno-oncology. The originator of this biosimilar is an agonist designed to cluster and activate CD40, with Fc engineering intended to enhance effector engagement. UniProt P25942 corresponds to human CD40, consistent with the card annotation.
