Background
This product is a research-grade biosimilar of Botensilimab, supplied as an unconjugated, non-therapeutic analog of the originator molecule and intended strictly for research use. It is built around the CTLA-4 target that Botensilimab engages and reproduces the antigen-binding specificity of the originator without being a clinical drug. It is provided as a human IgG1 (kappa) format and is manufactured to in-vivo research standards, including low endotoxin (research grade less than 1 EU/mg; ultra-low grade less than 0.5 EU/mg), with bulk milligram-to-gram quantities available for larger studies. Botensilimab is notable among anti-CTLA-4 agents for its Fc-enhanced design, which is intended to strengthen Fc-gamma-receptor engagement relative to first-generation CTLA-4 blockers; a biosimilar analog is useful as a reference or comparator reagent in binding, blockade, and effector-function work. Because this is a biosimilar rather than the therapeutic product, it is positioned for functional and in-vitro characterization, assay development, and use as a benchmarking control alongside other checkpoint-directed reagents rather than for any clinical or diagnostic application. CTLA-4 (CD152; UniProt P16410) is an inhibitory immune-checkpoint receptor expressed on activated conventional T cells and constitutively on regulatory T cells. It shares the ligands CD80 (B7-1) and CD86 (B7-2) with the costimulatory receptor CD28 but binds them with substantially higher affinity, allowing CTLA-4 to outcompete CD28 and dampen the costimulatory signal required for full T-cell activation. It also removes CD80/CD86 from antigen-presenting cells by transendocytosis, further limiting costimulation. By raising the activation threshold, CTLA-4 maintains peripheral tolerance and restrains autoimmunity. Blocking CTLA-4 releases this brake and can enhance T-cell priming and antitumor responses; Fc-enhanced anti-CTLA-4 antibodies are additionally thought to promote depletion of intratumoral regulatory T cells through Fc-gamma-receptor-mediated effector functions, which is the biology the Botensilimab molecule is designed to exploit.
