Background
This is a research-grade, unconjugated recombinant analog of zalutumumab, a fully human IgG1 kappa antibody directed against the epidermal growth factor receptor (EGFR/HER1). It is supplied strictly for research use only and is not the clinical therapeutic; it is not intended for human or veterinary use. The molecule reproduces the antigen-binding specificity of the originator against human EGFR, making it a convenient tool for studying receptor engagement, ligand-competition, and downstream signaling in cell-based systems. Because it is built on a human IgG1 backbone, it is useful for interrogating Fc-mediated effector functions such as antibody-dependent cellular cytotoxicity (ADCC) in vitro. Typical applications include positive and reference controls in EGFR binding and neutralisation assays, benchmarking of biosimilar or novel anti-EGFR candidates, flow cytometry and immunoassay standardisation, and as a building block for antibody-drug-conjugate (ADC) or engineering workflows. It is offered as low-endotoxin, bulk-scale material (research grade typically <1 EU/mg, with ultra-low <0.5 EU/mg options), supporting reproducible in-vitro and preclinical experimentation at milligram-to-gram scale. EGFR (encoded by EGFR, UniProt P00533; also called HER1/ErbB1) is a single-pass transmembrane receptor tyrosine kinase and a founding member of the ErbB family. Ligands such as EGF, TGF-alpha, amphiregulin, and epiregulin bind the extracellular domain, driving receptor homo- or heterodimerisation with other ErbB members (HER2, HER3, HER4). Dimerisation activates the intracellular kinase domain, triggering autophosphorylation and recruitment of adaptors that fire the RAS-RAF-MAPK, PI3K-AKT, and JAK-STAT pathways, which govern proliferation, survival, migration, and differentiation. EGFR is overexpressed, amplified, or mutated in many epithelial cancers, including head-and-neck squamous cell carcinoma, colorectal, and non-small-cell lung cancer, making it a validated oncology target. Zalutumumab binds the receptor ectodomain to block ligand-induced activation and can additionally engage Fc-dependent effector mechanisms.
