ichor.bio's Demupitamab Biosimilar (ICH5473) is an unconjugated, non-therapeutic research-grade analog of the originator antibody demupitamab, built around the epidermal growth factor receptor (EGFR, UniProt P00533). It is a human IgG1, kappa molecule intended strictly for research use, not for diagnostic or therapeutic application in humans or animals. As a biosimilar reagent it reproduces the target-binding specificity of the originator drug, providing investigators with a consistent, well-characterised tool for interrogating EGFR biology, benchmarking anti-EGFR binders, and serving as a positive control or reference standard in comparability, epitope-competition, and mechanism-of-action studies. EGFR (epidermal growth factor receptor; ErbB1/HER1, UniProt P00533) is a single-pass transmembrane receptor tyrosine kinase of the ErbB family. Ligand binding (EGF, TGF-alpha, amphiregulin and others) drives receptor homo- or heterodimerisation with other ErbB members, autophosphorylation of the intracellular kinase domain, and activation of downstream RAS-RAF-MEK-ERK, PI3K-AKT, and STAT signalling that governs proliferation, survival, migration, and differentiation. EGFR is frequently overexpressed, amplified, or mutated across epithelial malignancies including non-small-cell lung, colorectal, and head-and-neck cancers, where aberrant signalling promotes tumour growth and therapy resistance. Its extracellular domain is a validated target for antibody-based intervention, and the receptor is a central node in oncology and receptor-signalling research, making anti-EGFR reagents valuable for studying ligand-blocking, dimerisation, internalisation, and antibody-dependent effector mechanisms.
