Background
This product is an unconjugated, non-therapeutic recombinant analog of the anti-EGFR antibody modotuximab, supplied for research use only. It is a full-length human IgG1/kappa molecule engineered to recognize the human epidermal growth factor receptor (EGFR/ErbB1, UniProt P00533), providing a defined, reproducible reagent for studying EGFR biology without the regulatory and cost constraints of a clinical drug. Because it reproduces the binding specificity of the originator while being produced as a bulk research material, it is well suited to in-vitro and preclinical work: receptor-binding and blocking assays, competition and epitope-mapping studies, flow cytometry and immunoassay development, and as an isotype-matched positive control or benchmark against other anti-EGFR reagents. The IgG1 backbone also makes it useful for effector-function studies (ADCC/CDC) and as a starting scaffold for antibody-drug-conjugate or bispecific development. It is offered at low-endotoxin, research-grade purity (typically less than 1 EU/mg, with ultra-low-endotoxin options under 0.5 EU/mg) and in bulk milligram-to-gram quantities to support assay scale-up and standardization. It is not the clinical drug and is not for human or veterinary use. EGFR (epidermal growth factor receptor; ErbB1/HER1, UniProt P00533) is a single-pass transmembrane receptor tyrosine kinase and the founding member of the ErbB family. Ligands such as EGF, TGF-alpha, amphiregulin, and epiregulin bind the extracellular region and drive receptor homodimerization or heterodimerization with HER2, HER3, and HER4. Dimerization activates the intracellular kinase domain, leading to autophosphorylation of C-terminal tyrosines and recruitment of adaptors that trigger the RAS-RAF-MAPK, PI3K-AKT, and STAT pathways, promoting proliferation, survival, migration, and differentiation. EGFR is central to normal epithelial development but is frequently overexpressed, amplified, or mutated in cancers including colorectal, head and neck, lung, and glioblastoma, making it a heavily validated therapeutic and research target. Antibodies against its extracellular domain can block ligand binding and receptor activation.
