ICH5378 is a research-grade biosimilar of Becotatug, supplied as an unconjugated, non-therapeutic analog built around the same epidermal growth factor receptor (EGFR) target as the originator antibody. The parent molecule is a fully human IgG1-kappa monoclonal antibody directed against the extracellular domain of human EGFR. This research-grade material is not a clinical drug and is intended strictly for research use only. It is manufactured to low-endotoxin specifications (research grade, less than 1 EU/mg) and is available in bulk milligram-to-gram quantities, making it suitable as an in-vitro standard, positive control, or benchmarking reagent in EGFR-focused workflows. Because it reproduces the target-binding characteristics of the originator without the therapeutic payload or clinical formulation, it is well matched to functional assays comparing EGFR-directed binders. Epidermal growth factor receptor (EGFR, UniProt P00533; also known as ErbB1/HER1) is a receptor tyrosine kinase of the ErbB family. Ligand binding, by EGF, TGF-alpha, and related growth factors, drives receptor dimerisation and autophosphorylation of intracellular tyrosine residues, activating downstream RAS-RAF-MAPK, PI3K-AKT, and STAT signalling that governs cell proliferation, survival, migration, and differentiation. EGFR is expressed across many epithelial tissues, and its overexpression, amplification, or activating mutation is a well-established driver in numerous solid tumours, including nasopharyngeal, colorectal, head and neck, and non-small-cell lung cancers. This central role makes EGFR one of the most extensively targeted antigens in oncology, addressed by monoclonal antibodies against its extracellular domain and by small-molecule kinase inhibitors, and a frequent subject of binding, blocking, and receptor-internalisation studies.
