Background
This product is a research-grade biosimilar of ravulizumab, supplied as an unconjugated, non-therapeutic analog built around the complement component C5 target for research use only. It is not the clinical drug and is not intended for diagnostic or therapeutic use. Ravulizumab is a long-acting, humanized monoclonal antibody engineered from eculizumab with amino acid substitutions in the Fc and complementarity-determining regions that promote pH-dependent dissociation of C5 within the acidic endosome and FcRn-mediated antibody recycling, substantially extending serum half-life. The molecule carries an engineered IgG2/IgG4 hybrid framework with the M428L/N434S half-life-extending mutations reflected in the stated isotype. As a research reagent it provides a well-characterized, sequence-defined tool for studying terminal complement inhibition, C5 binding, and pathway blockade in controlled in-vitro systems. It is offered at low endotoxin, in-vivo research-grade standards and in bulk milligram-to-gram quantities suitable for functional assays, benchmarking, and assay development. Following activation of the classical, lectin, or alternative pathways, C5 convertases cleave C5 into C5a and C5b. C5a is a potent anaphylatoxin and chemoattractant that drives inflammation, neutrophil activation, and vascular permeability, while C5b nucleates assembly of the C5b-9 membrane attack complex (MAC), which forms lytic pores in target-cell membranes. Dysregulated terminal complement activity underlies conditions such as paroxysmal nocturnal hemoglobinuria and atypical hemolytic uremic syndrome. Antibodies that bind C5 and block its cleavage prevent generation of both C5a and the MAC, sparing upstream opsonization and immune-complex clearance. Ravulizumab binds C5 with pH-sensitive kinetics, enabling target release and sustained inhibition. C5 is a human protein, making it the relevant species context for this reagent.
